Получение длительно культивируемой эмбриогенной культуры Papaver rupifragum Boiss. & Reut. и ее цитологическое и биохимическое исследование

Аннотация
Впервые получена эмбриогенная культура мака скалоломного Papaver rupifragum Boiss. & Reut., которая была инициирована на корнях проростков, выращиваемых из семян на МС-среде с добавлением 1мг/л ИМК. Последующее поддержание эмбриогенной культуры осуществляли на безгормональной MС-среде. Длительное культивирование и сохранение эмбриогенной способности культуры (более 5 лет) поддерживалось благодаря циклам адвентивного эмбриоидогенеза, включающим образование каллуса на предсуществующих эмбриоидах и индукцию из их субповерхностных клеток новых эмбриоидов. В связи с этим полученная культура P. rupifragum может рассматриваться как дифференцированная культура, в которой стадия каллуса является промежуточным этапом развития. Установлено, что поверхность вновь образуемых эмбриоидов покрыта поверхностной сетью экстраклеточного матрикса, сформированной полисахаридами, липидами и терпенами. Гистологический анализ показал, что для эмбриогенной культуры P. rupifragum характерно образование комплексов слитых эмбриоидов (КСЭ), возникающих, предположительно, либо в результате кливажной полиэмбрионии, либо при синхронном развитии близлежащих эмбриоидов. Изучение гистологии и ультраструктуры КСЭ выявило, что слияние эмбриоидов обусловлено нарушениями в формировании эпидермиса и кутикулы. Гистохимические исследования установили, что эмбриогенная культура P. rupifragum синтезирует и накапливает триацилглицериды, полисахариды, фенольные соединения (ФС), терпены и алкалоиды. Показано, что количественный и качественный состав ФС и алкалоидов культуры P. rupifragum зависел от возраста культуры и ее дифференцировки, регулируемой условиями выращивания (свет, темнота). Дифференцированная эмбриогенная культура P. rupifragum сохраняет способность к формированию эмбриоидов на безгормональной МС-среде в течение длительного времени культивирования и может являться основой для дальнейшей разработки биотехнологических методов получения лекарственных соединений для косметологии и фармакологии.
